Description
The differential interference contrast (DIC) device is designed for studying transparent, low-contrast specimens that are difficult or impossible to observe using standard bright-field microscopy. This technique enhances image resolution and clarity through optical methods rather than chemical staining, while also providing additional structural information about the specimen.
The DIC method creates a pseudo-relief effect, giving the image an apparent three-dimensional appearance. The color and intensity variations in the image depend on changes in the refractive index and the thickness differences between adjacent areas of the specimen. Although the image appears three-dimensional, it does not represent the true physical geometry of the sample. Instead, contrast is based on optical thickness, which means the technique is not suitable for measuring actual height or depth.
DIC and phase-contrast microscopy complement each other. While both methods produce similar contrast images, DIC does not generate the diffraction halos commonly seen in phase-contrast images. These halos, caused by differences in refractive index between the specimen and its surrounding medium, often obscure fine structural details at object boundaries. DIC microscopy avoids this issue, allowing clearer visualization of edges and internal features.
The differential interference contrast system consists of a DIC condenser, a polarization filter, and DIC sliders.
DIC condenser
The DIC condenser is installed in place of the standard bright-field condenser. Its rotating turret contains multiple DIC prisms as well as an open slot used during bright-field observation. The appropriate prism is selected depending on the objective in use.
DIC sliders
Each DIC slider is mounted in a dedicated slot above the revolving nosepiece and is used together with a specific objective. The slider contains a DIC prism and an analyzer. The MAGUS DIC290 system includes two sliders: one for 10× and 20× objectives, and another for 40× and 100× objectives. Turning the slider adjustment knob moves the DIC prism laterally, allowing fine control of image contrast.
Polarization filter
For DIC operation, light from the illuminator must pass through a polarization filter, which is installed on the collector.
Recommended objectives
Objectives are not included with the MAGUS DIC290 device. DIC microscopy requires very precise alignment of optical components: the lower DIC prism must be positioned relative to the condenser’s front focal plane, and the upper DIC prism relative to the rear focal plane of the objective. The system is specifically designed for use with the MAGUS Bio 290T microscope, and only compatible or supplied objectives should be used. Objectives with higher numerical aperture, such as MAGUS FL S-APO60 PlanF models, are recommended. A higher aperture improves optical sectioning, allowing thin focal planes within thick specimens to be visualized, and also increases overall resolving power.
