Description
The epi-fluorescence attachment enables the use of the fluorescence technique, which relies on the ability of certain substances to emit light when excited by light of a specific wavelength. Some specimens display natural autofluorescence, while others require treatment with fluorochromes to produce a fluorescent signal.
Design and optical configuration
The attachment is installed in the optical path between the microscope stand and the head. Inside the unit is a five-position rotating turret containing four fluorescence filters with different wavelength ranges: broad-band blue (B), green (G), ultraviolet (UV), and narrow-band red (R). The desired filter is selected by rotating the turret ring. One turret position is dedicated to brightfield observation. Side knobs allow adjustment of the field diaphragm and aperture diaphragm openings.
Excitation light source
Excitation is provided by LEDs. These LEDs reach the user-selected brightness instantly, generate minimal heat even during extended operation, and can be switched off immediately when required. This makes it easy to switch between brightfield and fluorescence techniques. The LED service life is approximately 50,000 hours. LED-based fluorescence is recommended for higher-education environments because it is safe, reliable, and requires minimal maintenance.
Recommended configuration and optical performance
For fluorescence observations, the MAGUS Epi-FL05 LED attachment should be used with the MAGUS PlanF S-Apo fluorescent objective set. These plan semi-apochromatic fluorescent objectives improve resolving power and significantly reduce chromatic and spherical aberrations. As a result, images viewed through the eyepieces or captured with a digital camera are brighter, cleaner, and higher in contrast compared with images obtained using standard plan achromatic objectives.
The high numerical aperture of fluorescent objectives increases resolution but reduces depth of field. For transmitted-light observations, it is therefore recommended to use the standard objectives supplied with the microscope, as they provide a greater depth of field.
