Description
The epi-fluorescence attachment is designed for fluorescence microscopy. This technique is based on the ability of certain substances to emit light when excited by illumination at a specific wavelength. Some specimens show natural autofluorescence, while others require staining with fluorochromes to produce a fluorescent signal.
The attachment is installed in the optical path between the microscope head and the stand. Inside the unit is a four-position rotating turret equipped with three narrow-band fluorescence filter sets for the fluorochromes DAPI, FITC, and TRITC. The selected filter is brought into the optical path by rotating the control ring. One position of the turret is reserved for standard brightfield observation.
Excitation light source
Excitation is provided by LED light sources. The LEDs reach the selected brightness instantly, generate minimal heat even during extended operation, and can be switched off immediately when needed. This allows fast and convenient switching between brightfield and fluorescence observation. The LED lifetime is approximately 50,000 hours. For educational use, LED-based fluorescence is recommended because it is safe, reliable, and requires very little maintenance.
Objectives for fluorescence
To perform fluorescence microscopy, the MAGUS Epi-FL04 LED attachment is used together with the MAGUS PlanF S-Apo fluorescent objective set. Plan semi-apochromatic fluorescent objectives increase resolving power and reduce chromatic and spherical aberrations. Compared with standard plan achromatic objectives, they produce brighter images with higher contrast and improved clarity, both through the eyepieces and when using a digital camera.
Fluorescent objectives typically have a high numerical aperture, which improves resolution but reduces depth of field. For transmitted-light observation, it is therefore recommended to use the standard objectives supplied with the microscope, as they provide a greater depth of field.
