Description

The epi-fluorescence attachment is designed for fluorescence microscopy. This technique is based on the ability of certain substances to emit light when excited by illumination at a specific wavelength. Some specimens naturally exhibit autofluorescence, while others require treatment with fluorochromes to produce a visible fluorescent signal.

The attachment is installed in the optical path between the microscope head and the stand. Inside the unit is a six-position rotating turret fitted with fluorescence filters. Up to five fluorescence filter cubes can be installed in the turret at the same time, with the selection defined by the user. The desired filter is introduced into the optical path by rotating the control ring. One turret position is reserved for standard brightfield observation. Side-mounted knobs are used to adjust the field diaphragm and aperture diaphragm openings.

The available fluorescence filter cube sets include filters covering different wavelength ranges: broadband blue (B), green (G), ultraviolet (UV), violet (V), narrow-band red (R), as well as narrow-band filters designed specifically for the DAPI, FITC, and TRITC fluorochromes.

Excitation light source

Fluorescence excitation is provided by a mercury lamp. This light source offers high brightness and a broad wavelength spectrum with distinct intensity peaks, allowing effective use with a wide range of fluorescent dyes. The mercury lamp is housed within a dedicated lamphouse that dissipates heat efficiently, preventing the lamp from overheating during operation.

Objectives for fluorescence

For fluorescence microscopy, the MAGUS Epi-FL06 attachment is used together with the MAGUS PlanF S-Apo fluorescent objective set. Plan semi-apochromatic fluorescent objectives improve resolving power and significantly reduce chromatic and spherical aberrations. Compared with standard plan achromatic objectives, they deliver images that are brighter, cleaner, and higher in contrast, both through the eyepieces and when viewed with a digital camera.

Fluorescent objectives typically feature a high numerical aperture, which enhances resolution but reduces depth of field. For transmitted-light observations, it is therefore recommended to use standard objectives from the microscope set, as they provide a greater depth of field.

Compatibility

This epi-fluorescence attachment is compatible with the MAGUS Bio 290T biological microscope.
